Hamster kidney cells were transformed by circular or linear forms of the complete BK virus (BKV) genome and by a 3.7 kilobase fragment (fragment E) of BKV DNA, obtained by sequential digestion of BKV DNA with EcoRI and HhaI. The different species of BKV DNA showed a comparable transformation efficiency. Analysis by blot hybridization of the arrangement of viral DNA sequences in a cloned cell line transformed by BKV DNA fragment E showed the presence of eight viral integrations into the cellular DNA, six of which contained the entire early region of BKV DNA. Large T antigen (Ag), small t Ag and the non viral Tau antigen were detected in transformed cells by immunoprecipitation. The pattern of integration of viral sequences was constant over a high number of generations and large T Ag was always detected in transformed cells at various passage levels. These results are discussed in terms of the requirement of the entire early region for induction and maintenance of transformation by BKV....
