Five temperature-sensitive mutants of type 1 herpes simplex virus (HSV) have been isolated from BUdR-treated, HSV-infected Hep-2 cell cultures. Complementation experiments separated the mutants into 3 nonoverlapping groups. Preliminary phenotypic characterization of the mutants, at permissive (34°) and non-permissive (39°) temperature, indicated that: the mutant D10 in complementation group I did not replicate its deoxyribonicleic acid (DNA-) at 39° and, according to temperature-shift experiments, a temperature-sensitive event occurred before the sixth hour post-infection. The mutant F3 (group II) replicated viral DNA (DNA+) at 39° and temperature-shift experiments indicated that a temperature-sensitive event occurred before the ninth hour post-infection. Both D10 and F3 have been found to differ from wild-type in thermal stability at 45° and in their purified virion protein patterns, suggesting that they might possess altered structural proteins.
