The HIV-1 transactivator Tat is produced very early after infection and is key to the virus life cycle and to AIDS pathogenesis. A Tat-specific immune response in humans or monkeys (Mks) correlates with nonprogression to AIDS. Our recent studies indicate that native Tat activates DC functions and drives Th-1 type immune responses that are key to virus control. Further, vaccination of cynomolgus Mks with biologically active HIV-1 Tat protein or DNA is safe, immunogenic and contains primary infection with the SHIV89.6P.
