In this project a colloidal preparation in the form of lecithin/chitosan nanoparticles is proposed to optimize TAM oral absorption, reducing the variability in bioavailability observed with tablets. The objectives were to study such nanoparticles loaded with tamoxifen (TAM-LCN) for their physico-chemical properties as well as for cytotoxicity and cell uptake in vitro on cell lines. Moreover, ex-vivo experiments with excised rat intestinal mucosa allowed investigating TAM permeation across the mucosa when formulated in nanoparticles. The contribution of mucoadhesion to drug permeation was assessed.
